Evaluation of the Oral Toxicity Profile of a Nigerian Polyherbal Mixture (Oroki Herbal Mixture) In Rats.
The present study is an acute and subchronic toxicological evaluation of a Nigerian Polyherbal Mixture, Oroki Herbal Mixture, in female Wistar rats for 14 days and 90 days, respectively. Acute oral toxicity was conducted using the limit dose test of Up-And-Down Procedure at 5000 mg/kg of Oroki Herbal Mixture using the OECD Acute Oral Toxicity Testing 425 guidelines. For the subchronic oral toxicity test, twenty nulliparous female Wistar rats weighing between 120-150 g were divided into Groups I-IV that were orally treated with 10ml/kg of distilled water and 82 mg/kg, 410 mg/kg and 2050 mg/kg of Oroki Herbal Mixture, respectively, for 90 days. Thereafter, treated rats were humanely sacrificed under inhaled diethyl ether anesthesia and blood samples were collected for biochemical and hematological analyses and tissues harvested for histological assessment. Results showed that single oral treatment of rats with 5000 mg/kg body weight of Oroki Herbal Mixture extract produced no lethality in the three sequentially treated rats following acute oral toxicity testing with an estimated LD50 value of
